Colony PCR is an easy technique for quick screening of transformants and hence is used in many recombinant laboratories. However the major disadvantage is occurence of false positives. Though efforts have been done before to address this problem, but solution limits this technique by mandatory requirement of flanking vector oligos, which many a times is not possible. This work imparts universatility by removing this limitation and provides a quick method devoid of false positives for any cloning vector.
